FOXO4-DRI

FOXO4-DRI is a D-amino acid retro-inverso peptide derived from the FOXO4 transcription factor, engineered to penetrate cells and disrupt the interaction between FOXO4 and the tumor suppressor p53 — a binding interaction that is upregulated in senescent cells to sequester p53 and prevent the initiation of apoptosis, enabling senescent cell survival and the perpetuation of the senescence-associated secretory phenotype (SASP).

By competitively inhibiting this FOXO4-p53 interaction, FOXO4-DRI restores p53’s pro-apoptotic function specifically in senescent cells, triggering their selective clearance without affecting healthy proliferating or quiescent cell populations in preclinical models.
